    Abstract:  Living cells contain complex, analog circuits that regulate the rate at which each gene produces its product. The kinetic properties of these circuits enable cells to respond to changes in their environments and thus to survive, reproduce, and compete.  In this talk, I will describe work aimed at the efficient reconstruction of quantitative, kinetic models of gene regulatory networks.

    Quantitative modeling of gene regulatory networks can be roughly divided into a discovery phase and a refinement phase. In the discovery phase, the “wiring diagram” of a gene regulatory circuit is mapped, producing a rough quantitative model as a side effect. The refinement phase involves isolating components of the circuit through genetic engineering, varying the strengths of input signals, and quantitatively measuring the outputs. I will describe efforts to advance the methodology for both the discovery and refinement phases, as well as potential applications in biofuels, infectious disease, and metabolic disease.
